Resuscitation of soil microbiota after > 70-years of desiccation

Abstract The response pattern of soil bacterial communities to drought events has been characterized from short-term laboratory desiccation and in naturally arid soils, but the resistance bacteria persistently long periods remains largely unknown. This study assessed abundance composition 24 historical soils air-dried for more than 70 years, by quantification high-throughput sequencing analyses 16S rRNA genes. All contained measurable abundances varying 103 108 gene copies g?1 contrasting community compositions were observed with different land histories, suggesting that detected indigenous field. A 28-day rewetting incubation significantly increased indicating some these still alive quickly resuscitated. Among all identified taxa, Paenibacillus, Cohnella two unclassified Bacillales genera within phylum Firmicutes showed growth highest number (?12 soils), whereas Tumebacillus, Alicyclobacillus Brevibacillus displayed strongest activity (>1000-fold increase abundance) following rewet. Some Actinobacteria Proteobacteria also relatively high rewet, tolerance prolonged is a common trait across phylogenetically divergent microbes. present thus confirmed value desiccated detecting difference bacteria, provided evidence strong adaptation long-term extreme osmotic change.
